Pushing Forward With Zika Vaccines

چکیده

Approximately 70 years ago, new flavivirus was discovered in the Zika forest of Uganda (Dick et al., 1952). The eponymous virus is an arthropod-borne human pathogen that caused infrequent infections with relatively mild illness in Africa and received relatively little attention until recently. Over the last several years the virus has spread into Southeast Asia, across the Pacific and, in 2015, into Brazil causing larger and larger outbreaks. Most infected individuals (~80%) are asymptomatic, while those who develop clinical disease display short-lived symptoms including: rash, fever, arthralgia, and conjunctivitis (Barzon et al., 2016). Unfortunately, the recent outbreaks have corresponded with increased rates of severe fetal neurological defects, fetal malformations, and Guillain-Barre syndrome and subsequent studies have causally linked Zika virus to these serious clinical outcomes (Cao-Lormeau et al., 2016; Johansson et al., 2016).More recently, outbreaks have been reported in over 60 countries, with well over 1 million cases in, Brazil alone, leading theWHO to declare Zika a global public health emergency. Zika virus has been demonstrated to spread through sex and transfusionwith blood produces, and to be transmitted from themother to the fetus. The development of a safe and effective vaccine against Zika virus is a public health priority, however several challenges stand in our way. First, relatively little is known about Zika virus biology or immunity. Fortunately, other flaviviruses such as yellow fever virus, dengue virus, and Japanese encephalitis virus have been well studied, and have existing vaccines with a wealth of immunogenicity and efficacy data which can serve as starting model systems as Zika-focused research efforts come online. Second, the need to protect women who are pregnant or are

Zika Virus: Recent Advances towards the Development of Vaccines and Therapeutics

Zika is a rapidly emerging public health threat. Although clinical infection is frequently mild, significant neurological manifestations have been demonstrated in infants born to Zika virus (ZIKV) infected mothers. Due to the substantial ramifications of intrauterine infection, effective counter-measures are urgently needed. In order to develop effective anti-ZIKV vaccines and therapeutics, imp...

WHO reveals its shopping list for weapons against Zika.

If viruses had ears, Zika virus would be in bad shape after listening to a WHO official describing the armamentarium the world is getting ready to launch against it. “Based on our experience with research and development during the west Africa Ebola epidemic, WHO’s [R&D] response is proceeding very quickly for Zika”, Marie-Paule Kieny, WHO Assistant Director-General, Health Systems and Innovati...

Preventative Vaccines for Zika Virus Outbreak: Preliminary Evaluation

Since it emerged in Brazil in May 2015, the mosquito-borne Zika virus (ZIKV) has raised global concern due to its association with a significant rise in the number of infants born with microcephaly and neurological disorders such as Guillain-Barré syndrome. We developed prototype subunit and adenoviral-based Zika vaccines encoding the extracellular portion of the ZIKV envelope gene (E) fused to...

Development of Zika Virus Vaccines

Zika virus (ZIKV) is a mosquito-borne flavivirus that emerged as a global threat following the most recent outbreak in Brazil in 2015. ZIKV infection of pregnant women is associated with fetal abnormalities such as microcephaly, and infection of adults can lead to Guillain-Barré syndrome, an autoimmune disease characterized by neurological deficits. Although there are currently licensed vaccine...
